--it can be virus or it can be hardware problem or it can be pen drive or you could be disabled the usb port problem
1. using updated antivirus you can remove that virus.
2. for hardware problem, it usb cable inside the system may become loose check it out and check you pen drive.
3. Enable the usb port by following steps
3.a. right click the my computer and select manage, this in turn opens new window
3.b. in that select Device manage
3.c. right click on the usb port and check out whether it is enabled
