Hi Pete,
Please open Disk Management and search for your pendrive. It is possible that you need to assign a drive letter to it so that it can become accessible from My Computer. When you are in Disk Management, locate the pendrive and right click on it. Select assign letter. Please confirm that your pendrive has a proper file format.
Furthermore, I recommend you to test the pendrive on another PC confirm that it is working normally.
