=SUM(H5:N5)-O5
Enter the formula in cell P5.

Glad I could help.
Now, for your next query:-
If I have understood you correctly, then in cell S5, enter:-
=R5
and in cell S6 enter:-
=SUM(S5+R6)
I would be interested to know why you are using the IMSUM function. Are you adding complex numbers from P5 & Q5 in R5?
If so, I would like that other forum Contributors join this thread as complex number functions such as IMSUM (addition) or IMSUB (subtraction) don't come up that often on forums and for me it is still a very grey area (actually, it has been a grey area for me since High School!!!).
