Well, the drive has malfunctioned or become corrupt. I'd recommend you to try connecting it into different system and see if the system would able to read it.
You can also try following steps mentioned on below threads and see if that would help:

If nothing works, using recovery software would be your last resort. Once you get your files back, you can format the USB and start using it from scratch again.
Try Recuva or TestDisk (Pre Experience required).
